A study of mixing behaviour in disposable 2D bags used in cross-flow filtration

The time it takes to mix fluids in disposable bags is a key issue for many process developers. This study demonstrates recirculation of 1 cP and 5 cP viscous solutions in 50 L, 2-dimensional (2D) pillow bags using a tubing loop and peristaltic pump. This method is particularly applicable to cross flow filtration. The results show that good mixing in relatively short time can be achieved when sufficiently high recirculation flow rates are used. Many times well- mixed solutions are attained in three minutes or less.

AKTA ready system

ÄKTA ready is a liquid chromatography system built for process scale-up and manufacturing (Fig 1). It is part of the ReadyToProcess platform, a suite of plug-and-play, ready-to-use solutions for bioprocessing. The system operates with ready-to-use, disposable flow paths that are available separately in High and Low Flow Kits. As a benefit, the need for cleaning between products/batches is eliminated and no development and validation of cleaning procedures is required. Replacing flow paths between batches is fast, and when used together with ReadyToProcess columns, the risk for cross-contamination is removed.

An intensified perfusion one-step seed process with high-density cell banks

This work describes how the use of perfusion in the seed culture expansion process, in combination with the use of high-density cell banks, drastically can reduce processing time, simplify operations, and maximize equipment utilization (Fig 1). A high-density cell bank was created using a method that does not require centrifugation. One vial from this cell bank was used to inoculate a bioreactor culture that, when operated in perfusion mode, enabled a one-step seed culture process that could be used to seed a 2000 L production bioreactor.

Concentration and diafiltration of cell-derived, live influenza virus using 750 C hollow fiber filter cartridge

This application note describes the performance of GE Healthcare Life Sciences 750 C hollow fiber filter cartridge in concentration and diafiltration (buffer exchange) of cell culture-derived, live influenza virus. The 750 C cross flow filter is designed for cross flow filtration (concentration, impurity removal, and buffer exchange) of influenza virus and other entities of similar size. In this study, the influenza harvest was clarified using a normal flow filtration step, and then concentrated and diafiltered using the 750 C filter cartridge. Comparative experiments were performed with 500 C hollow fiber filter cartridge to evaluate differences in impurity removal capacity and flow characteristics. Intermediate and final fractions were analyzed for purity and yield of the target product. The well-defined molecular weight cut-off of the filter membrane allows separation of virus from impurities. Feed concentration and buffer exchange can be performed sequentially using the same equipment. The results show that both 500 C and 750 C hollow fiber filters enable achieving high virus yields (> 80%) and similar host cell protein (HCP) removal. However, the more open structure of the 750 C filter membrane resulted in a 1.5 to 2 order of magnitude higher removal of host cell DNA (hcDNA) compared with the 500 C filter.

Downstream scale-up purification of influenza virus using single-use bioprocessing equipment

This application note describes a case study where live influenza virus was purified using single-use bioprocessing technology. The GE Healthcare ReadyToProcess™ single-use platform is a flexible manufacturing platform which facilitates easy handling of virus solutions and helps improve productivity. As model system for a live, attenuated influenza virus vaccine, a A/H1N1/Solomon Island strain produced in Vero cells was used. Total process time for the downstream purification was 3 days and the overall scale-up process resulted in production of influenza virus with the same yield and purity as obtained in the laboratory scale.

Efficient, high-titer monoclonal antibody production in a fed-batch process using single-use stirred tank and rocking bioreactor systems

This application note shows the feasibility of monoclonal antibody (MAb) production in fed-batch processes using the single-use Xcellerex™ XDR-200 stirred-tank and ReadyToProcess WAVE™ 25 rocking bioreactor systems. Cell expansion was performed in a 15 L rocking bioreactor culture to seed a 200 L stirred-tank bioreactor culture. To compare the performance of ReadyToProcess WAVE 25 with that of XDR-200, 7 L of the 200 L culture was transferred to a parallel ReadyToProcess WAVE 25 system to a total culture volume of 10 L. MAb yields from the parallel productions were 5.0 and 4.9 g/L for XDR-200 and ReadyToProcess WAVE 25, respectively. The metabolite profiles were very similar in both bioreactor cultures. Together, the ReadyToProcess WAVE 25 and Xcellerex XDR bioreactor systems provide a powerful solution for robust, scalable production of recombinant proteins in a single- use format. In addition, the ReadyToProcess WAVE 25 is a versatile tool that can be used for both seed-train applications and process development.
