Growth of high-cell density microbial cultures in a single-use fermentor system

Here we show the performance of a single-use Xcellerex™ XDR-50 MO fermentor system when used in high-cell density cultivations of E. coli producing a domain antibody (Dab) and of modified Pseudomonas fluorecsens (P. fluorescens) producing a monoclonal antibody (MAb) (Fig 1). Both the achieved microbe densities and product yield were shown to be consistent with the performance of conventional stainless steel and glass bioreactor systems.

Optimization of product quality and yield in high-performing fed-batch CHO cell cultures

In this poster, we summarize data from studies of product quality of a human monoclonal antibody (IgG1) produced from Chinese hamster ovary (CHO) cells cultured in fed-batch mode. The effect of feed rate, pH, and temperature on cell growth and IgG production and quality was investigated with regard to glycan profile and charge variants. All studied parameters had a significant impact on both cell growth and productivity. The glycan profile and charge variant distribution could be altered by shifts in culture temperature or feed rate, whereas culture pH had less impact.

The studies summarized here increase the understanding of the impact of process conditions on cell culture performance and product quality.
