Optimizing a serum-free medium for high-density growth and efficient transient transfection of multiple HEK293 cell lines

Frequently in the biopharmaceutical industry, transient gene expression (TGE) is used to produce recombinant proteins for purposes such as biophysical characterization and clinical evaluation. These techniques require the rapid introduction of the gene of interest into human embryonic kidney (HEK293) cells. Often, this is done by complexing the recombinant DNA with a transfection reagent. The complexing process and subsequent introduction into the cells is frequently inhibited or extinguished by medium components, which has led to the use of very lean serum-free media for transfection and a different medium for production. Here we describe the use of a design of experiments (DoE) mixture design and high-throughput screening (HTS) techniques allowing for the development of a complete, serum-free medium formulation intended for both growth and transient transfection. The results show that very high transfection efficiencies and protein titers were achieved in the formulation, while using conventional transfection reagents and techniques such as Mr 25 000 linear polyethylenimine (PEI) and liposome-based transfection methods.

ReadyToProcess Columns

ReadyToProcess chromatography columns are validated high-performance bioprocessing columns that are supplied prepacked and ready for use. The columns are available with a range of BioProcess™ media (resins) in four different sizes (Fig 1). Standardized column formats allow off-the-shelf availability for short delivery lead times. ReadyToProcess columns are closed units and the design allows easy disposal after completed production, enabling flexibility in operations.

ReadyToProcess WAVE 25

The single-use ReadyToProcess WAVE 25 bioreactor system is a reliable and intuitive cell culture device for working volumes up to 25 L (Fig 1). The system is based on a well- known rocking technology that provides mixing and aeration to the culture, which is maintained in an inflated, disposable Cellbag™ bioreactor cultivation chamber.
