Scale-up of adherent Vero cells grown on Cytodex™ microcarriers using single-use bioprocessing equipment

In cell culture-based vaccine production, scale-up of adherent cells is challenging. This study shows a process for scaling up adherent Vero cells from static cell factories to influenza production at 50 L scale using WAVE Bioreactor™ systems and ReadyToProcess™ single- use bioprocessing equipment. Vero cells were grown to high cell density on Cytodex microcarriers in 10 L working volume. The cells were detached with trypsin and used to seed a 50 L production culture with the same microcarrier concentration. The cells were allowed to reattach and grow on the new microcarriers in a larger Cellbag™ bioreactor chamber. Cells were subsequently infected with influenza virus. The results show a repeatable scale- up procedure. The cell growth was similar in the 10 L seed cultures as in the 50 L production cultures in three consecutive experiments, with an obtained hemagglutinin (HA) concentration of approximately 12 μg/mL and a virus titer of 109 virus particles/mL.

Significant time-savings with simplified cell culturing using ReadyToProcess Wave 25

With today’s strict time frames, the demands for automation and ease of use in bioprocessing are rapidly increasing. The ReadyToProcess WAVE 25 bioreactor system was developed with the goal of keeping the simplicity of single-use rocking bioreactor systems, while enhancing system features with time-saving automation technologies (Fig 1). This application note describes how ReadyToProcess WAVE 25 can help reduce hands-on time as much as 25% in fed-batch culturing, while delivering excellent culture performance. For an optimized perfusion application, the use of intelligent control software enabled accurate regulation of process parameters. Over 100 × 106 viable cells/mL was easily achieved in seven days.

Simplified and reliable protein production from an insect cell baculovirus expression vector system (BEVS) using single-use ReadyToProcess WAVE bioreactor system

For applications such as research studies and preclinical trials, quick production of small amounts of recombinant proteins is often required. Protein production in insect cells using the baculovirus expression vector system (BEVS) is commonly used for such applications, as insect cells are capable of most post-translational modifications required for a fully functional and soluble product and the genetic engineering steps are straightforward. In this application note, we describe how ReadyToProcess WAVE 25 bioreactor system can simplify operations in insect cell applications, while providing stable culture conditions though accurate process control. This single- use system is easy to use and can minimize hands-on time compared with culturing in shaker flasks. Further, the enhanced control capabilities allow for a repeatable process outcome with reliable data to support conclusions for future decision making.

Utilization of high-throughput screening in the development of HyCell CHO medium

The Chinese hamster ovary (CHO) cell line is used extensively in the manufacture of recombinant human therapeutics. The nutritional requirements of this cell line vary specific to the individual recombinant clones isolated for this purpose. This property often necessitates the development of a clone-specific production medium to achieve optimal productivity in a bioprocess. Traditionally, such medium development efforts have primarily incorporated shake flask cultures, which limit the number of conditions, the number of replicates, and the ability to incorporate advanced design of experiments (DoE) statistical methods. In order to overcome these limitations, we investigated the use of various high- throughput screening (HTS) equipment in the development and testing of a medium designed for multiple CHO clones. HTS allowed for the inclusion of more individual cell lines, increased component factor-level and mixture design treatments, and all with a higher number of replicates yet less labor- intensive methods. Nearly all developmental work was performed at the 96-deep-well scale, with total culture volume at just over 1 mL. Subsequent scale-up and comparison of promising treatments, at shake flask scale, revealed exceptionally similar trends in both cell density and product quantity, validating the plate level design approach. Optimized medium developed with this approach demonstrates consistently higher protein yield across multiple clones. Comparisons of results at both scales are presented, along with a DoE approach to mixture design.
